Adenovirus Titration

We offer two methods to determine the infectious titer of an adenovirus stock.

  1. TCID50 method: Cell monolayers are seeded in multi-well plates and are infected with a serial dilution of the virus suspension to be titrated. Positive wells are counted generally from 12 to 18 days after the infection.  This is the most accurate and precise method.
  2. Plaque assay: Cell monolayers are seeded in tissue culture dishes.  They are infected with a serial dilution of the virus suspension to be titrated, and covered with agar. Plaques are counted generally from 12 to 18 days after the infection.

Applications

  • Determination of the concentration of infectious units (IU) in a crude or purified virus stock.

  •  A complete report describing the work performed, including the raw data and the determination of the titer

 Important Note on Handling Adenovirus

The use of adenovirus as a genetic vector requires adequate containment equipment and practices. Biosafety Level 2 (BL2) is appropriate for most customers; however, for some, use of BL3 or higher level containment facility may be required. PLEASE NOTE, O.D.260 Inc. will not carry out projects that require BL3 (or above) containment.
