10 reactions The Cosmid Construction Kit -2 contains most of the reagents necessary to construct AdenoQuick2.0 cosmids:
- The SfiI restriction enzyme is used to digest the 4 shuttle plasmids before ligating them to each other.
- OD101 is the E. coli strain used to construct and amplify adenovirus cosmid DNAs.
- Maltose and MgSO4 are added to the E. coli culture to induce expression of the receptor for phage λ
- The λ packaging extract is used to package the DNA ligation products into the phage λ heads.
- The SM medium is used to stop the packaging reaction at the end of the 30-minutes incubation period.
- The Amp/Kan antibiotic mix will be used to select for colonies containing the adenovirus cosmid.
The reagents that the user needs to provide are:
- T4 DNA ligase
- Chloroform
- LB Lennox (liquid and petri dishes)
The SfiI restriction enzyme must be stored at - 20 ºC. The lambda packaging extract must be stored at - 70 ºC.
